ABSTRACT

1,2,4-triazine ring is a prominent structural motif found in numerous natural and synthetic biologically active compounds. This prompted us to synthesize new fused 1,2,4-triazine derivatives and evaluate their antimicrobial and anticancer activities. The first step is to prepare 5-Benzylidene-3-(p-cholorophenyl)-2-aminothiocarbonyl-1,2,4-triazine (2) which was carried out via the condensation of oxazolinone (1) with thiosemicarbazide. The reaction of (2) with bomomethyl aryl ketones, ethyl chloroacetate and acetic anhydride give the corresponding fused 1,2,4-triazine derivatives (3, 4 and 9). Treatment of 4 with acetic anhydride and aromatic aldehydes yielded the corresponding acetyl, diacetyl derivatives (6 and 7) and also triazino-1,2,4-traizine derivatives (8). All synthesized 1,2,4-triazine derivatives were characterized by elemental analysis, infrared, mass and 1H-NMR spectroscopic techniques. All the synthesized compounds were screened for their antimicrobial activity. Also the cytotoxicity and antitumor activity of some prepared 1,2,4-triazine derivatives were tested against MCF-7 cell line. Results revealed that Triazolo-1,2,4-triazine and 1,2,4-triazine derivatives (9 and 2) were found to exhibit potent anticancer activity better than other compounds.
